Can I link my square account deposit to an American Express Account to deposit instead of a regular bank account?

Could you share some more details about the American Express account? For example is it a pre-paid debit card account? Or is it an American Express savings account?

Currently you can link a bank account that accepts deposits and withdrawals. Most banks with brick and mortar locations are supported. 

Pre-paid debit cards cannot be linked as a deposit method.

Yes you can link a savings account, as long as it is a transactional account that supports deposits as well as debits (in case of chargebacks).
